Despite the loss, the Panthers clinched their fourth straight playoff berth and fifth consecutive trip to the postseason.
Florida led 1-0 on a goal from Vladimir Tarasenko and 2-1 on a goal from Sasha Barkov.
The Islanders scored twice to end the second period and that was the final scoring.
Anthony Stolarz made 21 saves in the loss.
Florida will play host to the Detroit Red Wings on Saturday at 12:30 p.m.
